چکیده

We investigated effect of docosahexaenoic acid on lipoprotein synthesis and secretion by cultured eel hepatocytes. When eel hepatocytes were incubated with 1 mM docosahexaenoic acid (DHA) at 28 degrees C for 24 h, triacylglycerol (TG), free cholesterol (FC), and cholesteryl ester (CE) syntheses from 14C-acetate increased 9.7-, 1.6-, and 8.5-fold, respectively, and the specific activity of TG was twofold that of control. These results indicate the stimulative effect of DHA on lipid synthesis. However, the radioactivity of the lipoprotein secreted by DHA-treated cells incubated with 14C-acetate and 3H-leucine reduced to 55 and 60%, respectively, compared to that by control cells, but DHA did not affect the incorporation of 3H-leucine into other secreted protein. Furthermore, the amount of the secreted lipoprotein per 24 h by DHA-treated cells also reduced to 17%. These results show that DHA inhibits the secretion of lipoprotein. Intracellular lipids such as TG, PL, and TC increased by 4.8, 1.6, and 1.8 times, respectively, in DHA-treated cells. The remarkable increase in TG in DHA-treated cells seems to be due to the stimulative effect of DHA on TG synthesis and an inhibitory effect of DHA on lipoprotein secretion, since about 70% of the secreted lipoprotein consists of TG. The chemical composition and apoprotein profile of the lipoprotein secreted by DHA-treated and control cells were almost the same. These results suggest that DHA inhibits the assembly or secretion of the lipoprotein in eel hepatocytes.

Studies on Eel Liver Functions Using Perfused Liver and Primary Cultured Hepatocytes

Perfused eel livers, isolated eel hepatocytes, and cultured eel hepatocytes are used to investigate eel liver functions such as gluconeogenesis, glycogen synthesis, and lipoprotein synthesis and methods for preparation are described. A novel phosphoenolpyruvate (PEP) synthesis pathway from pyruvate in gluconeogenesis in eel liver was elucidated and PEP synthesis pathways in eel, rat, and pigeon...

Effect of specific binding of high density lipoprotein to eel hepatocytes on their secretion of lipoprotein.

Specific binding of eel serum high-density lipoprotein (HDL) to eel hepatocytes was demonstrated by using a synthesized fluorescent lipophilic dye. HDL binding was inhibited by the addition of unlabeled HDL. The binding of HDL to the hepatocytes was saturated at concentrations over 100 micrograms HDL protein/ml and Kd value was 20 micrograms HDL protein/ml. A fluorescent photomicrograph of the ...

HEPATOMA McARDLE-RH7777 CELLS HAVE THE SAME RESPONSE AS NORMAL RAT HEPATOCYTES TO BOTH DIBUTYRYL-cAMP AND ANTICALMODULINW-7

The effects of cAMP-analogue dibutyryl-cAMP and anticalmodulin W-7 were studied on de novo synthesis and secretion of lipids in cultures of hepatoma McArdle RH7777 cells and normal rat hepatocytes. Dibutyryl-cAMP and W -7 separately caused a significant decrease in the secretion of de novo synthesized triacyl [3H]glycerol in both cultures of McArdle cells and rat hepatocytes. The inhibito...

Characterization of lipoprotein secreted by cultured eel hepatocytes and its comparison with serum lipoproteins.

The lipoprotein secreted by cultured eel hepatocytes was fractionated by density gradient ultracentrifugation and compared with eel serum lipoproteins. Eel hepatocytes were cultured for 7 to 10 days as a monolayer in Williams' medium E containing 5% fetal bovine serum and 0.16 microM insulin on a dish precoated with fibronectin of horse serum. The only lipoprotein secreted by eel hepatocytes wa...

The effects and interaction between cAMP-analogue dibutyryl-cAMP and calmodulin antagonists were investigated on de novo synthesis and secretion of lipids in cultures of hepatoma McArdle-RH7777 cells and normal rat hepatocytes. Dibutyryl cAMP caused a significant decrease in the secretion of de novo synthesized triacyl [3H] glycerol in both cultures of McArdle cells and rat hepatocytes. The ...
